Description

The Nisin controlled gene expression system (NICE®), developed at NIZO Food Research, NL, is a fully food-grade system used for controlled gene expression in Lactococcus lactis. The NICE® system is advantageous in that it does not contain endotoxins, inclusion bodies, spores or extracellular proteases, and the tightly controlled gene expression allows the production of toxic proteins to take place. With this system, fermentation, scale-up and downstream processing are simple.

The NICE®pNZ9530 Lactococcus lactis nisRnisK vector, part of the NICE® expression system, is a low copy plasmid with pAMΒ1 origin of replication that carries the nisR and nisK genes. It is used for cloning in Lactococcus strains and in strains of other lactic acid bacteria genera that do not have the regulatory genes integrated into the chromosome. In this case, a two-plasmid system is used for nisin induced expression, such as pNZ9530 (nisRnisK) and pNZ8150 (+insert).
