The ELISA-VIDITEST anti-HHV-6 IgG assay kit is intended for the serological diagnosis of diseases associated with HHV–6 infection, such as exanthema subitum, acute respiratory illnesses, diarrhea with fever and febrile seizures in infants, heterophile antibody-negative infectious mononucleosis in children, and interstitial pneumonia, encephalitis, meningitis, hepatitis and aplastic anemia in immunodeficient patients. The presence of IgG anti-HHV-6 antibody reveals the immune status of the patient. A significant rise in anti-HHV-6 IgG antibodies in paired serum samples, taken in the acute and convalescent phases of the infection, is indicative of the active infection. The test does not differentiate between HHV-6 subtype A and B.
This assay is a solid-phase immunoanalytical test that detects IgG antibodies to HHV-6 in human serum. The strips are coated with native HHV-6 antigen. If present in the sera tested, the anti-HHV-6 antibodies bind to the immobilized antigens. The antibodies in complexes with antigens are later recognized by animal anti-human IgG antibodies labeled with horseradish peroxidase. The attached labeled antibodies are revealed by an enzymatic reaction with a chromogenic substrate. Negative sera do not react. A mild change in color, if present, may be attributed to the reaction background.

- Prepare reagents and samples
- Dispense 100 µl/well standards, control and samples; incubate 60 min. at room temperature; wash 4 times (250 µl/well), aspirate
- Dispense 100 µl/well Px-conjugate; incubate 60 min. at room temperature; wash 4 times (250 µl/well), aspirate
- Dispense 100 µl/well TMB-O substrate; incubate 20 min. at room temperature
- Dispense 100 µl/well Stop solution
- Read the absorbance at 450 / 620-690 nm within 20 minutes
