CloneTracker™ XP 10M 3' Barcode Libraries

This off-the-shelf, ready-to-use barcode libraries label individual cells with unique barcodes that can be detected by genomic DNA amplification and in RNA sequencing analysis.The barcodes are incorporated into the UTR region of an RNA transcript. They are encapsulated in VSV-g pseudotyped lentiviral particles and can transduce and integrate into the host cell genome of most mammalian cells. When transduced, individual cells integrate a single barcode into their genomic DNA. The barcode sequences replicate as the host cell divides and are passed on to the daughter cell. After selection, treatment, or differentiation, the genomic DNA can be extracted, amplified, and sequences to identify and quantify barcodes present or expressed and quantified using RNA-Seq.For RNA-Seq, the 5’UTR is compatible with SMART-based cDNA synthesis and the 3’UTR is compatible with oligo-dT cDNA synthesis protocols.
