DriverMap™ AIR Spike-In Controls

Cellecta's DriverMap™ Adaptive Immune Receptor (AIR) RNA Spike-In Controls are synthetic messenger RNA (mRNA) reference standards designed to evaluate, calibrate, and standardize T-cell receptor (TCR) and B-cell receptor (BCR) repertoire sequencing assays. These controls support assessment of assay sensitivity, specificity, quantitative accuracy, sequencing errors, and sample-to-sample contamination, helping researchers improve the reliability and reproducibility of immune clonotype profiling.

The controls consist of 48 synthetic BCR constructs representing 16 receptor isoforms and 39 synthetic TCR constructs representing 13 receptor isoforms. Each isoform includes three distinguishable CDR3 sequence variants within a nearly full-length V(D)J-C receptor structure. Collectively, the controls represent all seven immune receptor chains: TRA, TRB, TRG, and TRD for TCRs, and IGH, IGK, and IGL for BCRs.

Designed for use with multiplex reverse transcription PCR (RT-PCR) and 5′ rapid amplification of cDNA ends (5′-RACE/SMART) PCR-based sequencing methods, the controls are compatible with both commercial and laboratory-developed adaptive immune receptor profiling workflows.

DriverMap AIR RNA Spike-In Controls are available as Premixed Controls or Triplex Isoform Pools. Premixed Controls contain defined concentrations of synthetic receptor variants for evaluating detection sensitivity, quantitative linearity, and sequencing accuracy. Triplex Isoform Pools contain individual receptor isoform pools with distinguishable CDR3 variants, providing flexibility for assay validation, sample quality assessment, and cross-contamination studies.

BCR Standards

Fig 1: 48 BCR mRNA spike-in constructs for IGH, IGK and IGL chains are shown. Each construct has three variations in the CDR3 region that differ by three nucleotides in a fixed position.
