This vector is a ready-to-use mammalian expression vector intended for investigation of the human Mis12 complex. It encodes a fusion protein of the fluorescent protein EGFP and human Mis12. These fusion proteins can be used in modern fluorescence microscopy techniques (FRET, FRAP, FCS, RICS, FCCS, and F3H) to determine localization, binding, dynamics, protein-protein interactions and protein-protein proximities. The cipher in the vector name indicates the linker length of the fusion proteins.
The Mis12 complex includes four proteins. A multi-protein complex called the kinetochore binds at each centromere. The inner kinetochore directly binds to the centromeric chromatin during the whole cell cycle. During mitosis, further proteins of the outer kinetochore bind to the inner kinetochore and establish the stable link between the centromeric chromatin and the microtubuli. This mechanical connection is regulated by proteins of the mitotic checkpoint. The Mis12 complex is a link between the inner and outer kinetochore.
