DriverMap™ AIR-RNA Profiling Assays
Cellecta's DriverMap™ AIR-RNA Assays provide highly sensitive and comprehensive profiling of T-cell receptor (TCR) and B-cell receptor (BCR) repertoires from total RNA. Using multiplex reverse transcription PCR (RT-PCR) and next-generation sequencing (NGS), these assays enable researchers to identify and quantify expressed immune receptor clonotypes, assess repertoire diversity, and investigate clonal expansion and changes in T- and B-cell populations.
The assays are available for CDR3 or full-length variable-region profiling. CDR3 assays utilize experimentally validated multiplex primer sets targeting the FR3 variable region and conserved constant regions of TCR/BCR transcripts. Full-length assays incorporate additional FR1-targeting primers to characterize the complete variable region, including CDR1, CDR2, and CDR3. Depending on the selected kit, researchers can profile four TCR chains (TRA, TRB, TRD, TRG), three BCR chains (IGH, IGK, IGL), or combined TCR/BCR repertoires.
RNA-based profiling provides greater sensitivity for detecting low-abundance expressed clonotypes than DNA-based profiling, making it particularly valuable for studies of immune responses, repertoire diversity, and antigen-associated clonal changes. Cellecta's proprietary Validator Barcode (VBC) technology uses internal barcoded replicates to improve clonotype quantification and help identify amplification and sequencing errors.
The assays support RNA isolated from whole blood, peripheral blood mononuclear cells (PBMCs), immune cell fractions, tissues, and other biological specimens. FFPE samples may also be analyzed when RNA quality is sufficient.
Required Accessory: A compatible Universal Unique Dual Index (UDI) NGS Index Plate must be purchased separately for sequencing libraries prepared with DriverMap AIR-RNA assays.
