DriverMap™ Single-Cell AIR (scAIR) Chain Profiling Assays
Cellecta's DriverMap™ Single-Cell Adaptive Immune Receptor (scAIR) Chain Profiling Assays enable full-length sequencing and paired-chain identification of T-cell receptors (TCRs) or B-cell receptors (BCRs) from individually sorted or pipetted cells. Using a streamlined, 96-well plate-based multiplex RT-PCR and next-generation sequencing (NGS) workflow, these assays provide detailed information about immune receptor sequences, clonotypes, and receptor-chain pairing.
The assays characterize paired TCR alpha and beta chains (TRA/TRB) or BCR heavy and light chains (IGH/IGK/IGL), providing receptor information that cannot be reliably reconstructed from conventional bulk immune repertoire profiling alone.
Cells are deposited into indexed 96-well plates containing gene-specific reverse transcription primers. Following primer hybridization, samples are pooled into a single reaction tube for subsequent amplification and NGS library preparation. Well-specific barcodes preserve cell-of-origin information, while Cellecta's proprietary Validator Barcodes (VBCs) support quantitative analysis and help identify amplification and sequencing errors.
The workflow eliminates the need for separate RNA extraction, microfluidics platforms, or complex manual barcoding procedures. Assays can be performed using individually sorted cells or small pools of cells per well, supporting both single-cell receptor characterization and analysis of more abundant receptor-chain pairs within larger cell populations.
DriverMap scAIR Chain Profiling Assays are available for human, mouse, and hybrid human-mouse samples, with separate TCR and BCR configurations.
Required Accessory: A compatible Universal Unique Dual Index (UDI) NGS Index Plate must be purchased separately for sequencing libraries prepared with DriverMap scAIR assays.
