Description
These off-the-shelf, ready-to-use human genome-wide dual-sgRNA library is designed to activate gene expression. The CRISPRa Dual-sgRNA Library targets nearly 19,000 human protein-coding genes, with 5 sgRNAs per gene. The library uses the 5 sgRNAs in dual combinations across 5 constructs (for example, sgRNA 1 & 2, sgRNA 1 & 3, sgRNA 2 & 3, etc.). In addition, the dual-sgRNA format provides increased levels of gene activation compared with the single-sgRNA library.This is the only system that enables genome-wide gain-of-function screens to identify endogenous genes that alter phenotypes when activated rather than knocked out or turned off. Unlike a CRISPR knockout, the induction is reversible as it does not alter the underlying DNA structure. This library provides a tool for identifying candidate therapeutic targets involved in the regulation of growth, differentiation, disease pathogenesis, and other biological responses. Libraries are available as 200 ug plasmid or pre-packaged viral libraries at 2 x 10^8 TU and 1 x 10^9 TU, and in the following vector formats: UbiC-RFP-Puro, UbiC-Puro, andUbiC-RFP-Bleo.
- Designed to work with deactivated spCas9 (dCas9) complexed with an activator (dCas9-VPH) to activate gene expression
